Currently it seems totem can only handle DVD subtitles - and you can not select a file on your local computer that contains substitles like in format srt or ASS.
This makes totem useless for many videos.

You can right-click on a file in the playlist to add subtitles, and in 2.26 you'll be able to select subtitles via the "View" menu.
