Currently, GNOME Tweak Tool do not offer an option to customize desktop’s font face and size.

GNOME Tweak Tool needs an additional font selector button under “Fonts” section to edit dconf key org > gnome > nautilus > desktop > font, similar to the existing ones; or modify font face and size at the same time as the “default” font.
