No, it would be correctly stated as "system-wide settings" because the two words are being used together as one adjective.  Using separate words means that they are both separate adjectives describing settings, "system, wide settings" which is not what you mean; the settings are not both "system settings" and "wide settings", but "system-wide settings".  However, if not used as a combined adjective, do not hyphenate: contrast, for example "the oil-on-canvas painting" with "he used oil on canvas for his painting".  Of course, there are exceptions such as the word "very" which is usually not hyphenated.
