Since totem is able to read some .iso files, maybe it should add application/x-cd-image to the list of supported MIME types.

Not sure about that. For most people, ISO images won't be video DVDs, or VCDs, but just CD/DVD images.

A fix to bug 530651 would allow us to see "Open in Movie Player" when the ISO is mounted.

It looks like a better alternative indeed. It would also require some UI for mounting the ISO.
